The combined effect of hydroxyurea and x-rays on Chinese hamster cells in vitro.
نویسنده
چکیده
Hydroxyurea inhibits DNA synthesis in Chinese hamster cells and is selectively toxic for cells actually in DNA synthesis (i.e., in S phase) at the time of exposure to the drug. Synchronized cells in G1 inhibited from entering S by hydroxyurea show a complex response to X-radiation. Cells become steadily more sensitive for a period of about 4 hr and then recover about a normal G1 sensitivity in approximately a further 4 hr. They never achieve the high survival level normally found for uninhibited cells in late S, but they will reach this level quite rapidly if the hydroxyurea is removed prior to exposure. The sensitization is, therefore, completely reversible. Hydroxyurea has no effect if present only during exposure at room temperature. Additional sensitization is obtained if the drug is present after exposure which increases for a period of about 4 hr. "This postirradiation sensitization is independent of the time in the cycle at which irradiation takes place. These results with synchronized populations enable the effects of hydroxyurea on asynchronous populations to be predicted. The most sensitive condition occurs when irradiation is given about 4 hr after the administration of hydroxyurea and the drug is kept on subsequently. The results indicate that hydroxyurea in combination with X-rays can be a most effective toxic agent for Chinese hamster cells. If these results are applicable to in vivo situations, combination therapy with hydroxyurea given systemically and X-rays delivered locally to tumors may be most effective. However, in view of some of the problems in tumor control that cannot be evaluated in vitro, experiments with suitable animal tumor systems are needed.

Interphase Death of Chinese Hamster Ovary Cells Exposed to Accelerated Heavy Ions
Introduction: Heavy ions are nucleus of elements of iron, argon, carbon and neon that all carry positive electrical charges. For these particles to be useful in radiotherapy they need to accelerated to high energy by more than thousand mega volts. Also the cosmic environment is considered to be a complicated mixture of highly energetic photons and heavy ions such as iron. Therefore, the health ...

Background: Follicle stimulating hormone (FSH) is one of the most common and effective gonadotropin hormones used in infertility treatment. Production of FSH as a drug is affordable as long as it is done with minimal cost. FBS is the most expensive component in medium culture furthermore; it’s being controversial to cause unnecessary suffering for the unborn calf for collection their serum. In ...

Background: The demand for producing recombinant therapeutic proteins by mammalian cell lines, such as Chinese hamster ovary (CHO) cells, continues to grow. Significant achievements in process optimization including development of cell culture strategies for largescale and cost-effective production have been made. Fetal bovine serum (FBS) is an often essential growth supplement and yet most cos...
متن کاملInduction of cancer stem-like cells in A549 cells after exposure to carbon ions and X-rays
Background: Cancer stem-like cells (CSCs) play a crucial role in the initiation, progression, and recurrence of cancer. Evidence indicates that the high linear energy transfer (LET) carbon ion beam is more effective against CSCs than the conventional X-ray beam. Carbon ion radiotherapy is considered as a promising cancer strategy, however, information about whether, or not, new CSCs are induced...

Background: IFN-g is mostly secreted by activated CD4+ , CD8+ T cells and NK cells. This cytokine has immunomodulatory, anti-cancer and anti-microbial effects and is important for prophylaxis, diagnosis and treatment of chronic infections and cancers. Objective: The purpose of this study was to clone the full cDNA of human IFN-g and express it in CHO cell line. Methods: Lymphocytes from a healt...
